Abstract

The aim of the present study include two lines, the first one is to isolation and identification. The second line was included determination the role of silver nanoparticles with WSPAgs in stimulating immune responses against virulence Escherichia coli O157:H7in mice. To achieve the first goal, 150 sample of (diarrhea) were collected from human100 diarrheal sample and 50 sample from water of river using routine bacteriological isolation and identification. The result showed that among 100 human sample which equal (62 %),  were bacterial. Positive ,the result of water from Euphrates River in AL- Fallujah ( 50 sample) was 27 positive isolate which equal (54%) . The bacteria isolated from diarrheal samples of human and from sample of river water too. The second goal was achieved, by fifty white mice, average age 8 to 10 weeks, were divided randomly into five groups equally and treated as following, 1st group was immunized 1/P by 0.2ml of WSPAgs, protein concentration 0.5mg/ml, two doses, two weeks interval,2nd group was inoculated with 0.2ml of AgNPs , two doses ,two weeks interval , 3rd group was immunized with 0.2ml of mixed WSPAgs and silver nanoparticle with ratio 1:1 as in the 1st group,4th group was served as control positive and 5th group was served as control negative, at 28 day post immunization ,skin test was done ,using soluble sonicated Escherichia coli antigens. At 30 days post immunization, five animals from 1st, 2nd,3rd,5th groups were sacrificed and blood samples were collected in order to determine serum levels of IFN y and antibodies titers by ELISA, than 1st , 2nd , 3rd,4th group were inoculated with o.2ml of bacterial suspension containing 1X104 cfu/ml , I/P, 5th group was inoculated with 0.2ml of sterile normal saline. It is concluded that bacterial strains were associated with diarrhea of human E. coli O157:H7 that AgNPs can augment protective immunity against E. coli O157:H7 infection.
